pRha-RBS-BLS for controlled expression of beta-lactam synthetase
Our composite part was designed by combining pRha (Part: BBa_K914003), a Ribosome Binding Site (RBS) (Part: BBa_B0034) and BLS (Part: BBa_K2372002).
The promoter pRha is used to control the expression of the target gene.
The RBS is part of the mRNA and binds upstream at the 5’ End to the DNA. A RBSs task is to position the mRNAat the right place. The efficiency of the bonding determines how well the translation is accomplished.
β-lactam Synthetase (BLS) catalyzes the formation of a β-lactam ring in clavulanic acid, which is a β-lactamase inhibitor. β-Lactamase is believed to be one of the main reasons of antibiotic resistance. Clavulanic acid has proven effective against a wide range of antibiotic resistant organisms.
